Abstract

The invention provides construction of a recombinant adenovirus carrier for expressing mouse nerve growth factors. Genetic engineering technology is applied, mouse nerve growth factor genes are inserted into a downstream of a cytomegalovirus promoter of the carrier, recombined shuttle plasmids and virus framework plasmids are jointly converted into escherichia coli DH5a competence bacteria, and the recombinant adenovirus plasmid vector containing target genes are obtained. The adenovirus plasmid vector is used for transfection of 293T cells to obtain the recombinant adenovirus carrier with the mouse nerve growth factor genes, a constructed adenovirus carrier containing nerve growth factor (NGF) genes passes through diabetes mouse hearts and is used for transfection of the diabetes mouse hearts, heart sensory nerve conduction functions can be obviously improved, and glycuresis nerve ending lesion degree is lightened. The adenovirus carrier is applied to preparation of medicines for treating glycuresis combined coronary heart diseases.
本发明提供一种表达小鼠神经生长因子的重组腺病毒载体的构建,应用基因工程技术,将小鼠神经生长因子基因插入到载体巨细胞病毒启动子的下游,将重组的穿梭质粒与病毒骨架质粒共转化大肠杆菌DH5ɑ感受态细菌,获得含有目的基因的重组腺病毒质粒载体。将该腺病毒质粒载体转染293T细胞,获得带有小鼠神经生长因子基因的重组腺病毒载体,所构建的含NGF基因腺病毒载体,经过并将其转染到糖尿病小鼠心脏,可明显改善心脏感觉神经传导功能,减轻糖尿病神经末梢病变程度。所述腺病毒载体可在制备治疗糖尿病合并冠心病药物中的应用。
